Central nervous system (CNS) disorders are some of the most prevalent yet poorly treated illnesses. Achieving sufficient blood-brain barrier (BBB) penetration is one of the biggest challenges in the development of therapeutics for CNS disorders. For instance, according to an article published by National Center for Biotechnology Information, in January 2005, more than 98% of small molecular drugs and almost 100% of larger molecular drugs are ineffective in treating CNS diseases considering that BBB strictly limits the delivery of therapeutic agents to their target site in the CNS.
